Heterotopic Model

A heterotopic model is an experimental biology approach in which a tissue, organ, or graft is studied in an anatomical location different from its normal site, allowing researchers to separate its intrinsic behavior from local tissue influences. In transplantation studies, the graft is implanted at a non-native site and may be connected to the recipient’s blood supply, enabling it to remain viable while investigators monitor rejection, vascularization, growth, or function. These models support research on organ transplantation, immune responses, tissue repair, and developmental signaling, while enabling controlled comparisons with orthotopic models.

This study presents a pre-clinical heterotopic abdominal heart xenotransplantation model using α-Gal knockout (GTKO) pigs and macaques. The model incorporates optimized recipient selection, vascular anastomosis techniques, and perioperative management strategies. It is suitable for evaluating novel immunosuppressive agents and provides a convenient platform for advancing heart xenotransplantation.

The mouse heterotopic heart transplantation model has been proven by many investigators to be an important method for studying mechanisms of rejection and immune response. However, the techniques involved are still challenging. By modifying standard techniques we have had success with more than 1000 transplants.
