Histone Demethylase

Histone demethylases are enzymes that remove methyl groups from histone proteins, helping regulate how DNA is packaged and which genes are expressed. They act on specific methylated lysine residues through distinct catalytic mechanisms: LSD1-family enzymes use flavin adenine dinucleotide for oxidative demethylation, while JmjC-domain enzymes require iron and 2-oxoglutarate to hydroxylate the methyl group before its removal. By reshaping chromatin states, histone demethylases influence cell differentiation, development, DNA repair, and disease-associated gene regulation. Their selective activity makes them valuable subjects for studying epigenetic mechanisms and potential targets for therapies involving cancer and other disorders.
