Homologous Desensitization

Homologous desensitization is a biological process in which a cell selectively reduces its response to a receptor that has been persistently activated by its specific ligand. For many G protein-coupled receptors, ligand binding promotes phosphorylation by G protein-coupled receptor kinases, enabling arrestin binding, disruption of receptor-G protein coupling, and sometimes receptor internalization. This receptor-specific feedback limits excessive signaling while allowing other receptors and pathways to remain responsive. Studying homologous desensitization helps explain cellular adaptation and signal regulation and supports research on drug tolerance, receptor-targeted therapies, and the design of compounds with sustained or selective effects.
