Hplc Purification

High-performance liquid chromatography (HPLC) purification is a separation method that isolates and cleans chemical compounds from complex mixtures, making it valuable for analyzing and preparing biomolecules in biology. It works by pumping a liquid mobile phase through a column containing a stationary phase, where compounds interact differently with the column material and therefore elute at distinct retention times; collected fractions can then be evaluated for purity. In biological research, HPLC purification supports the isolation of peptides, proteins, nucleotides, metabolites, and pharmaceutical compounds, enabling structural analysis, functional studies, quality control, and reproducible downstream experiments.

Herein we report a tailored HPLC purification protocol that yields high-purity amyloid beta 42 (Aβ42) and amyloid beta 40 (Aβ40) peptides, capable of oligomer formation. Amyloid beta is a highly aggregation prone, hydrophobic peptide implicated in Alzheimer's disease. The amyloidogenic nature of the peptide makes its purification a challenge.
