Immature Embryos

Immature embryos are developing embryonic structures that have not yet completed the cellular differentiation, tissue organization, and maturation needed for independent growth. Their development depends on coordinated cell division, cell expansion, and lineage specification, regulated by genetic programs and signals from surrounding tissues, including nutrients and hormones. In biology, studying immature embryos reveals how body plans and specialized tissues arise during early development. Isolated embryos can also be maintained or grown under controlled culture conditions, supporting embryo rescue, developmental studies, and research on factors that influence viability, maturation, and subsequent growth.

Generation of Immature, Mature, and Tolerogenic Dendritic Cells from Monocytes

This video demonstrates the generation of immature, mature, and tolerogenic dendritic cells, or DCs, from human monocytes. The incubation of monocytes with growth factors and cytokines leads to the production of immature DCs. In response to immune regulatory molecules, immature DCs differentiate into tolerogenic and mature DCs.

Immature dendritic cells can be selectively differentiated into tolerogenic or mature dendritic cells to regulate the balance between immunity and tolerance. This work presents a means to generate from immature monocyte derived dendritic cells (moDCs), in vitro tolerogenic and mature moDCs that differ in metabolic phenotypes.

siRNA-Based Inhibition of Autophagy in Herpes Simplex Virus-Infected Immature Dendritic Cells

This video demonstrates a small interfering RNA (siRNA)-based technique to interfere with autophagic flux. A siRNA targeting FIP200 mRNA is introduced into monocyte-derived immature dendritic cells (iDCs) using electroporation, causing the knockdown of FIP200 expression and leading to the inhibition of nuclear lamin disruption via autophagy. Upon adding herpes simplex virus (HSV), the nuclear egress of viral progeny is inhibited due to the absence of lamin disruption via autophagic flux.
