Immunomodulatory Effects

Immunomodulatory effects are the changes that a substance, cell, or biological signal produces in immune system activity, including the enhancement, suppression, or regulation of immune responses. These effects arise when immunomodulatory factors influence immune-cell signaling, cytokine release, antigen recognition, or communication between innate and adaptive immune pathways. In biology, studying these mechanisms helps explain how organisms respond to infection, maintain tolerance to healthy tissues, and resolve inflammation. Characterizing immunomodulatory effects also supports research into vaccines, autoimmune disease, cancer, transplantation, and therapies designed to restore balanced immune function.

A Fluorescence-Based Assay Using Engineered Lymphocytes for Screening Immunomodulatory Compounds

This video demonstrates the screening of immunomodulatory compounds via fluorescent-based assay. Transgenic mouse-derived T lymphocytes with green fluorescent protein expression cassettes are treated with these compounds. The enhanced and reduced GFP signals enable the identification of the compounds' stimulatory and inhibitory effects, respectively.

This video demonstrates an in vitro assay to explore the immunomodulatory influence of mesenchymal stem cells (MSCs) on effector T cell proliferation. The assay includes co-culturing MSCs with PBMCs to induce the monocyte population to acquire an immunomodulatory phenotype, leading to the secretion of interleukin-10 (IL-10). Following incubation with activated CD4+ T cells, the induced monocytes effectively suppress T cell proliferation.

Rapid Quantification of Mitogen-induced Blastogenesis in T Lymphocytes for Identifying Immunomodulatory Drugs

T-lymphocyte mitogenesis is accompanied by blastogenic transformation, whereupon the cell volume enlarges before cell division. Here, we describe a method to quantify blastogenesis in T lymphocytes using an automated cell counter with the capability of measuring cell diameters.

The immunomodulatory properties of human mesenchymal stem cells (MSC) appear increasingly relevant for clinical application. Using a co-culture system of MSCs and peripheral blood leukocytes pre-stained with the fluorescent dye carboxyfluorescein succinimidyl ester (CFSE), we describe the in vitro assessment of MSC immunomodulation on effector leukocyte proliferation and specific subpopulations.
