Apical Surface Stimulation

Apical surface stimulation is the controlled application of a chemical, mechanical, or biological stimulus to the outward-facing membrane of a polarized epithelial cell. Because the apical membrane contains specialized receptors, ion channels, and transport proteins, stimulation can activate membrane signaling, alter ion fluxes, and trigger intracellular responses such as calcium elevation or changes in gene expression. In biology, this approach helps model how epithelia sense their luminal environment, including nutrients, pathogens, drugs, and physical forces. It is useful for studying epithelial transport, barrier function, host-microbe interactions, and tissue responses in cultured cells, organoids, and other experimental systems.

A step-by-step video protocol of apical resection is demonstrated in this study. Apical resection is a recently highlighted surgical approach in mammalian heart regeneration research. This study may promote the application of apical resection as a standard methodology in research into the mechanism underlying heart regeneration.

We introduce a novel method for the maintenance of human intestinal mucosa in culture and monitoring of the response to various types of stimuli over at least 24 hrs. With our method, the polarity of the tissue is maintained, allowing for a physiological stimulation via the apical route.

This protocol describes an apical-out necrotizing enterocolitis (NEC)-in-a-dish model utilizing small intestinal enteroids with reversed polarity, allowing access to the apical surface. We provide an immunofluorescent staining protocol to detect NEC-related epithelial disruption and a method to determine the viability of apical-out enteroids subjected to the NEC-in-a-dish protocol.
