In Vivo Transplantation

In vivo transplantation is the transfer of cells, tissues, or organs into a living recipient to restore function, study biological responses, or evaluate therapeutic potential. After implantation, the graft must interact with the host environment, establish adequate nutrient and blood supply, and either avoid or manage immune recognition that can cause rejection. In biology, this approach supports research on tissue regeneration, stem cell behavior, organ function, and disease progression, while providing a physiologically relevant setting for assessing graft survival, integration, and therapeutic effects. Findings from in vivo transplantation can guide the development of regenerative treatments and improve understanding of host-graft interactions.

Normothermic Ex Vivo Kidney Perfusion for the Preservation of Kidney Grafts prior to Transplantation

The severe organ shortage has resulted in increased use of marginal kidney grafts for transplantation. This has triggered interest in alternative storage methods, since marginal grafts especially tolerate cold storage poorly. The technique of normothermic ex vivo kidney perfusion (NEVKP) represents a novel preservation method for kidney grafts prior to transplantation.

Here, we present a protocol to induce tolerance in transplantation, and assess in vitro and in vivo the suppressive capacity of distinct cell subsets from the recipient and the immune status of the recipient toward donor or exogenous antigens.

GABAergic cortical interneuron progenitors disperse, develop and synaptically integrate into a host cortex after transplantation. These cells can be easily transduced before transplantation for in vivo studies of genetically modified GABAergic precursors. Here, we show viral labeling techniques to target specific interneuron subgroups using existing Cre lines and Cre-dependent reporters.
