Outer Membrane Isolation

Outer membrane isolation is a laboratory technique for separating the outer membrane of Gram-negative bacteria from other cellular components, enabling analysis of its structure and function. The process typically disrupts bacterial cells, removes cytoplasmic contents and inner membranes, and uses differential centrifugation or density-gradient ultracentrifugation to enrich the membrane fraction, which contains lipopolysaccharide, outer-membrane proteins, and associated molecules. Isolated membranes support studies of bacterial envelope architecture, permeability, host-pathogen interactions, antibiotic resistance, and vaccine targets. Careful fractionation and purity assessment are essential because contamination from inner membranes or soluble proteins can alter biochemical and proteomic results.

Here, we describe the purification of Legionella pneumophila (L. pneumophila) outer membrane vesicles (OMVs) from liquid cultures. These purified vesicles are then used for the treatment of macrophages to analyze their pro-inflammatory potential.
