Keratinocyte Fibroblast Coculture

Keratinocyte fibroblast coculture is an in vitro model that grows epidermal keratinocytes together with dermal fibroblasts to reproduce key cellular interactions in skin biology. Fibroblasts support keratinocyte behavior through paracrine signaling, secretion of extracellular matrix components, and release of growth factors, while keratinocytes can influence fibroblast activity through reciprocal signals. This interaction helps researchers study epidermal differentiation, skin barrier formation, wound repair, inflammation, and tissue remodeling under controlled conditions. Keratinocyte fibroblast coculture also supports development of engineered skin, evaluation of biomaterials, and testing of topical drugs or cosmetic ingredients, providing a more physiologically relevant system than either cell type alone.

An in vitro method to mimic in vivo epithelial differentiation is described. Many viruses target epithelial cells as part of their viral life cycle, and this method provides a means of examining virus:host interactions that more closely resembles that which occurs in vivo. This technique can be used with primary keratinocytes, established cell lines, as well as normal or diseased biopsy tissue.
