Two-kidney One-clip

Two-kidney one-clip is an experimental model of renovascular hypertension in which a partial constriction is applied to one renal artery while the opposite kidney remains normally perfused. Reduced blood flow to the clipped kidney stimulates renin release, activating the renin-angiotensin-aldosterone system and increasing vasoconstriction, sodium retention, and blood pressure. This Biology model helps researchers examine how renal perfusion, hormonal signaling, and kidney function contribute to hypertension. It is also used to evaluate cardiovascular and renal effects of sustained high blood pressure and to investigate potential antihypertensive therapies.

A modified 2 kidney 1 clip (2K1C) Goldblatt mouse model was developed using polyurethane tubing to initiate renal artery stenosis, inducing an increase in renin expression and kidney injury. Here, we describe a detailed procedure of preparing and placing the cuff onto the renal artery to generate a reproducible and consistent 2K1C mouse model.

Kidney Structure

The kidneys are two large bean-shaped organs located in the upper abdomen. They filter the blood several times a day to remove toxins and rebalance water and electrolytes of the circulatory system via the renal veins. The kidneys receive blood directly from the heart via the renal arteries. These arteries enter the kidney at the hilum, the concave surface of the bean, where they branch and divide into smaller vessels and capillaries. The renal cortex is the thick outer layer of the kidney. It...

The administration of drugs for recovery of kidney function requires control of the localization and distribution of the therapeutic compound. Here, we describe in detail a simple technique for intrarenal delivery of drugs in rats. This procedure may be easily performed with no mortality and high reproducibility.
