Larval Isolation

Larval isolation is the separation and recovery of larval-stage organisms from their surrounding culture, host tissue, or environmental sample for study or further rearing. The process typically combines careful collection with physical separation methods, such as transferring larvae by pipette or sieve and removing debris under controlled conditions that preserve viability and developmental state. Isolated larvae can then be identified, counted, cultured, imaged, or exposed to experimental conditions. In biology, this technique supports investigations of development, behavior, physiology, genetics, and environmental responses while enabling researchers to analyze larvae independently of mixed-stage populations.

Drosophila Larval IHC

2023

Immunohistochemistry (IHC) is a technique used to visualize the presence and location of proteins within tissues. Drosophila larvae are particularly amenable to IHC because of the ease with which they can be processed for staining. Additionally, the larvae are transparent, meaning that some tissues can be visualized without the need for dissection. In IHC, proteins are ultimately detected with antibodies that specifically bind to “epitopes” within the protein of interest. In order to preserve...

Source: Li, J., et. al., Development of a Larval Zebrafish Infection Model for Clostridioides difficile. J. Vis. Exp. (2020)This video demonstrates the isolation and culturing of Clostridioides difficile bacteria from the intestine of larval zebrafish. The infected intestine is dissected, homogenized, and incubated in selective anaerobic media containing antibiotics. These conditions suppress competing microbes and promote the growth of viable cells.
