Layer 2/3 Neurons

Layer 2/3 neurons are nerve cells located in the superficial layers of the cerebral cortex, where they help process and distribute information across cortical circuits. Many are excitatory pyramidal neurons that integrate synaptic inputs through their dendrites and send signals to nearby neurons and other cortical areas, supporting communication between local and distributed networks. Their connectivity and activity contribute to sensory processing, perception, learning, and experience-dependent plasticity. Studying Layer 2/3 neurons helps researchers explain how cortical information is transformed and shared, while also providing insight into circuit dysfunction associated with neurological and psychiatric disorders.

Here, we show how to analyze dendritic routing of Drosophila medulla neurons in columns and layers. The workflow includes a dual-view imaging technique to improve the image quality and computational tools for tracing, registering dendritic arbors to the reference column array and for analyzing the dendritic structures in 3D space.
