Layer Isolation Protocol

Layer Isolation Protocol is a biological method for separating distinct layers within a tissue, cultured sample, or other structured biological material so each compartment can be studied independently. The procedure typically preserves the sample’s organization while using controlled dissection, enzymatic treatment, mechanical separation, or selective collection to isolate layers according to their position and physical or cellular properties. Isolated layers can then undergo microscopy, cell culture, molecular analysis, or functional assays. This approach helps researchers compare layer-specific composition, cell behavior, signaling, and responses, supporting studies of tissue organization, development, disease mechanisms, and tissue engineering.

Neutrophils are among the first cells to arrive on the site of inflammatory immune response, and their functions and mechanisms have been studied extensively in vitro. We demonstrate a standard density gradient separation method to isolate human neutrophils from whole blood using commercially available separation media.

The method described has been specifically adapted for studies of the viral etiology of tonsil inflammation known as tonsillitis.

Layer-by-layer Collagen Deposition in Microfluidic Devices for Microtissue Stabilization

The creation of functional microtissues within microfluidic devices requires the stabilization of cell phenotypes by adapting traditional cell culture techniques to the limited spatial dimensions in microdevices. Modification of collagen allows the layer-by-layer deposition of ultrathin collagen assemblies that can stabilize primary cells, such as hepatocytes, as microfluidic tissue models.
