Interleukin-2 Secretion

Interleukin-2 (IL-2) secretion is the regulated release of a cytokine that coordinates immune-cell growth, survival, and activation, making it central to adaptive immunity. When a T cell recognizes antigen through its receptor and receives appropriate costimulatory signals, intracellular signaling activates IL2 gene transcription; newly synthesized IL-2 is then released and binds IL-2 receptors on the producing cell and nearby lymphocytes. This autocrine and paracrine signaling promotes T-cell proliferation and supports the development and function of regulatory T cells and other immune populations. Measuring IL-2 secretion helps assess cellular immune responses, vaccine activity, immunotherapies, and immune-modulating treatments.

Functional Assessment of Antibody-Secreting B Cells Using ELISpot Assay

This video demonstrates the use of the ELISpot assay to assess the function of antibody-secreting B cells, ASCs. The antibodies secreted by the ASCs interact with the membrane-bound polyclonal antibody fragments, which are detected by the enzyme-conjugated IgG or IgM antibodies. The colored spot produced in each well indicates the presence of the specific antibody's isotype, which correlates with the B cells' secretory activity.
