Melanin Quantification

Melanin quantification is the measurement of melanin content in cells, tissues, or biological samples, providing a numerical assessment of pigmentation and melanogenic activity. In a common approach, melanin is extracted or solubilized from a sample, and its concentration is estimated by measuring light absorbance with spectrophotometry against a standard or calibration curve; results may be normalized to cell number or total protein. This method supports studies of melanocyte function, pigmentation disorders, skin biology, and responses to drugs or environmental stimuli. Reliable quantification helps researchers compare melanin production across experimental conditions and evaluate mechanisms that regulate pigment synthesis.

2025

A basic protocol for fabricating high-transconductance Melanin/PEDOT:PSS organic electrochemical transistors (OECTs) using spray coating is described. The method allows precise control over film deposition, thickness, and morphology, providing reproducible devices for sustainable and scalable bioelectronic applications.

Pharmacologic Induction of Epidermal Melanin and Protection Against Sunburn in a Humanized Mouse Model

Epidermal melanin is induced by topical application of forskolin in a murine model of the fair-skinned UV-sensitive human. Pharmacologic manipulation of cAMP levels in the skin and epidermal darkening strongly protect against UV-mediated inflammation (sunburn) as measured by the minimum erythematous dose (MED) assay.

Invertebrate Lifespan Quantification

2023

Many animals naturally stop growing upon reaching adulthood, after which they undergo aging or "senescence" until dying. The amount of time between an organism\'s birth and death is called its lifespan, which can be influenced by various biological and environmental factors. By exposing organisms to different growth conditions, scientists can better understand the factors affecting lifespan. Flies and worms are ideal organisms to perform such experiments, given their short generation time and...

Colorimetric Assay for Quantification of Lactate in Test Samples

2025

This video describes a colorimetric assay for lactate measurement in cell lysates by using the lactate dehydrogenase enzyme. This enzyme converts lactate into pyruvate, along with the conversion of NAD to NADH that reacts with tetrazolium dyes and gives a colored product that can be measured spectrophotometrically to quantitate the lactate.
