Membrane Trafficking

Membrane trafficking is the coordinated movement of proteins, lipids, and other cargo between membrane-bound compartments in eukaryotic cells, a process essential for maintaining cellular organization and communication. Cargo is selectively packaged into transport vesicles that bud from donor membranes, move through the cytoplasm, and fuse with specific target membranes through coat proteins, Rab GTPases, tethering factors, and SNARE proteins. This system supports secretion, endocytosis, organelle biogenesis, and recycling of membrane components. In biology research, studying membrane trafficking helps explain nutrient uptake, signal regulation, neuronal function, and how trafficking defects contribute to infection, genetic disease, and cancer.

Using an α-Bungarotoxin Binding Site Tag to Study GABA A Receptor Membrane Localization and Trafficking

Here we demonstrate the use of fluorescent Alexa dye coupled to α-bungarotoxin to measure GABA A receptor surface localization and endocytosis in hippocampal neurons. Through the use of constructs bearing a short extracellular tag that binds α-bungarotoxin, analysis of plasma membrane protein endocytic trafficking can be achieved.

Analysis of SCAP N-glycosylation and Trafficking in Human Cells

We describe a modified method for membrane fraction isolation from human cells and sample preparation for the detection of SCAP N-glycosylation and total protein by using western blot. We further introduce a GFP-labeling method to monitor SCAP trafficking using confocal microscopy. This protocol can be used in regular biology laboratories.

While the transport of cell surface proteins is relatively easily studied, visualizing the trafficking of intracellular proteins is much more difficult. Here, we use constructs incorporating photoactivatable GFP and demonstrate a method to accurately follow the amyloid precursor protein from the Golgi apparatus to down-stream compartments and follow its clearance.

Neuronal membrane trafficking dynamically controls plasma membrane protein availability and significantly impacts neurotransmission. To date, it has been challenging to measure neuronal endocytic trafficking in adult neurons. Here, we describe a highly effective, quantitative method to measure rapid changes in surface protein expression ex vivo in acute brain slices.
