Microrna Mimic Transfection

MicroRNA mimic transfection is a laboratory method for introducing synthetic, double-stranded RNA molecules into cells to reproduce or enhance the activity of a naturally occurring microRNA. After delivery, the mimic is incorporated into the RNA-induced silencing complex, where its guide strand directs recognition of complementary sequences in target messenger RNAs, reducing their stability or translation. In biology research, this approach helps investigators test microRNA function, identify regulated genes and pathways, and model changes in processes such as development, differentiation, and disease. It also supports evaluation of microRNA-based strategies for therapeutic development, although delivery efficiency and cellular toxicity require careful assessment.

MicroRNAs (miRNAs) are important regulators of gene expression and have been shown to play a role in numerous biological processes. To better understand miRNA-UTR interactions, we have created a genome-wide collection of 3 UTR luciferase reporters paired with a novel luciferase gene and assay reagent, the LightSwitch system.

We describe here a method for regulating picornavirus tropism by incorporating sequences complementary to specific microRNAs into the viral genome. This protocol can be adapted to all different classes of viruses with modifications based upon the length and nature of their life cycle.

Ectopic expression is one technique to elucidate the microRNAs role in brain development. However, targeting specific areas using in ovo electroporation is challenging. Here, we show an efficient way to selectively electroporate ventral and dorsal midbrain regions.
