Mouse Peritoneal Injection

Mouse peritoneal injection is a laboratory technique for delivering fluids, drugs, cells, or other experimental materials into the peritoneal cavity of a mouse, enabling controlled studies of absorption, treatment, and biological response. During the procedure, the animal is appropriately restrained and a needle is inserted through the abdominal wall into the cavity, where the administered material can distribute and cross the peritoneal lining for systemic or local effects. This route supports pharmacology, immunology, toxicology, and disease-modeling research, while careful technique helps reduce tissue injury, variability, and unintended organ puncture.

The peritoneal cavity in mammals contains different immune cell populations crucial for innate immune responses. An efficient isolation method is required for biochemical and functional analyses of these cells. Here we provide a comprehensive method for the isolation of peritoneal cavity cells in the mouse.

(2025)This video demonstrates the quantification of intracellular survival of a methicillin-resistant bacterial pathogen in mouse peritoneal macrophages. It outlines the steps involved in infection, extracellular bacterial clearance, host cell lysis, and colony enumeration to assess bacterial survival within host macrophages.
