Ms5 Stromal Cells

Ms5 stromal cells are an immortalized mouse bone marrow-derived cell line used to model the cellular microenvironment that supports blood-cell development. In co-culture systems, they form an adherent stromal layer that maintains hematopoietic cells through direct cell contact and the release of soluble signals, helping reproduce key features of the bone marrow niche in vitro. Researchers use Ms5 cells to study hematopoietic stem and progenitor cell maintenance, lineage differentiation, and interactions between stromal and blood-forming cells. These cultures provide a controllable platform for investigating mechanisms of hematopoiesis and evaluating how genetic, molecular, or pharmacological changes affect blood-cell production.

Generating Schwann Cell-Like Cells from Bone Marrow Stromal Cells

This video demonstrates a method for obtaining Schwann cell-like cells from multipotent bone marrow stromal cells (BMSCs). Initially, the BMSCs undergo hypoxia treatment, followed by directed differentiation into neuronal progenitor cells. Subsequently, specific media and culture conditions are employed to induce the transformation of these progenitor cells into Schwann cell-like cells.

The video demonstrates a method for investigating the impact of stromal cells on leukocyte recruitment. In a parallel plate flow chamber model, pro-inflammatory cytokines induce the expression of adhesion receptors by endothelial cells, facilitating neutrophil recruitment and migration. The cross-talk between stromal cells and endothelial cells diminishes neutrophil recruitment to the endothelial cells.
