Multiple Chamber System

A multiple chamber system is an experimental platform that organizes biological samples, cells, or tissues into two or more distinct compartments to study their interactions under controlled conditions. Chambers may be separated by permeable membranes, porous barriers, or microfluidic channels, allowing researchers to regulate contact, fluid exchange, molecular transport, and chemical gradients while keeping populations physically distinct. In biology, these systems support co-culture experiments, cell migration studies, barrier and transport assays, and investigations of signaling between tissues or cell types. By isolating variables and enabling parallel measurements, they improve the analysis of complex biological interactions and help model tissue interfaces more realistically than single-compartment cultures.

Light sheet-based fluorescence microscopy is the most valuable tool in developmental biology. A major issue in comparative studies is ambient variance. Our protocol describes an experimental framework for simultaneous live imaging of multiple specimens and, therefore, addresses this issue pro-actively.

Here, a workflow for the culture and gene expression analysis of endothelial cells under fluid shear stress is presented. Included is a physical arrangement for simultaneously housing and monitoring multiple flow chambers in a controlled environment and the use of an exogenous reference RNA for quantitative PCR.

Precise measurement of neurological and neuropsychological impairment and disability in multiple sclerosis is challenging. We report methodologic details on a new test, the Multiple Sclerosis Performance Test (MSPT). This new approach to the objective of quantification of MS related disability provides a computer-based platform for precise, valid measurement of MS severity.
