Mycobacterium Species

Mycobacterium species are rod-shaped bacteria distinguished by lipid-rich cell walls and diverse ecological, medical, and environmental roles. Their waxy walls contain mycolic acids, which reduce permeability and help cells retain carbol fuchsin during acid-fast staining, while also contributing to resistance against drying and some chemical stresses. The genus includes pathogenic species such as Mycobacterium tuberculosis, which causes tuberculosis, as well as nontuberculous species found in soil and water that can cause opportunistic infections. Studying their classification, physiology, and antibiotic susceptibility supports clinical diagnosis, treatment selection, public-health surveillance, and research on bacterial persistence and host-pathogen interactions.

Mycobacterium tuberculosis forms drug tolerant biofilms when cultured in certain conditions. Here we describe methods for culturing M. tuberculosis biofilms and determining the frequency of drug tolerant persisters. These protocols will be useful for further studies into the mechanisms of drug tolerance in M. tuberculosis.
