Origin Recognition

Origin recognition is the biological process by which a cell identifies sites on DNA where chromosome replication will begin, helping ensure accurate genome duplication. In eukaryotes, the origin recognition complex binds replication-origin DNA and recruits licensing proteins, including the MCM helicase, to assemble a pre-replication complex; cell-cycle controls activate these origins during S phase. This regulated process coordinates DNA unwinding, primer synthesis, and bidirectional replication while restricting each origin to one initiation event per cell cycle. Studying origin recognition clarifies genome stability and cell-cycle regulation, and helps explain how replication defects can contribute to disease.
