Pancreatic Lipase

Pancreatic lipase is a digestive enzyme that breaks down dietary triglycerides, making it essential for lipid digestion and the absorption of fat-soluble nutrients. Secreted by the pancreas into the small intestine, it acts at the oil-water interface and hydrolyzes ester bonds in triglycerides, primarily producing free fatty acids and 2-monoglycerides; colipase helps maintain this activity in the presence of bile salts. In biology and medicine, pancreatic lipase provides a model for enzyme catalysis and supports the evaluation of digestive function, pancreatic insufficiency, fat malabsorption, and treatments that modify lipid digestion.

This protocol explains a method for isolating pancreatic islets by first perfusing collagenase via Ampulla of Vater to digest the exocrine pancreatic tissue and a simplified density gradient centrifugation to purify the islets from mice.
