Pdms Microwells

PDMS microwells are arrays of microscale cavities molded in polydimethylsiloxane, an optically transparent and elastomeric material used to organize cells under controlled conditions. Their recessed geometry confines cells within individual wells, while surface properties and well dimensions influence cell attachment, aggregation, and the formation of three-dimensional structures. In biology, PDMS microwells support high-throughput generation of uniform cell spheroids, organoids, and other multicellular assemblies for studying development, cell interactions, and disease models. Because researchers can image and manipulate many standardized cultures in parallel, these platforms improve experimental consistency and enable applications in drug screening, tissue engineering, and regenerative research.

This manuscript introduces a robust method of fabricating concave microwells without the need for complex high-cost facilities. Using magnetic force, steel beads, and a through-hole array, several hundred microwells were formed in a 3 cm x 3 cm polydimethylsiloxane (PDMS) substrate.
