Peroxisomal Protein Import

Peroxisomal protein import is the process by which newly synthesized proteins are selectively transported from the cytosol into peroxisomes, organelles that support lipid metabolism, detoxification, and other essential functions. Most matrix proteins carry a peroxisomal targeting signal, such as PTS1 or PTS2, which is recognized by receptors including PEX5 or PEX7; these receptor-protein complexes dock at the peroxisomal membrane, deliver cargo through a transient import channel, and release the receptors for recycling. Studying this pathway clarifies how cells maintain organelle composition and function, while defects in import machinery can cause severe peroxisomal disorders and reveal mechanisms of human disease.

Here we describe a protocol to express proteins into protoplasts by using PEG-mediated transformation method. The method provides easy expression of proteins of interest, and efficient investigation of protein localization and the import process for various experimental conditions in vivo.
