Plant Tissue Clearing

Plant tissue clearing is a set of methods that makes opaque plant samples sufficiently transparent for three-dimensional imaging while preserving their internal organization. These methods typically remove or chemically reduce light-absorbing pigments, extract selected cellular components, and adjust the tissue’s refractive index to reduce light scattering; some protocols also label specific structures with fluorescent markers. Cleared tissues can then be examined by light-sheet, confocal, or other fluorescence microscopy to visualize cells, vascular networks, roots, leaves, and developing organs in their spatial context. The approach supports plant anatomy, developmental biology, physiology, and disease research by revealing complex structures that conventional sectioning may disrupt.
