Protozoa Culture In Vitro

Protozoa culture in vitro is the laboratory maintenance and propagation of unicellular eukaryotes outside their natural hosts or environments, enabling controlled study of their biology. Researchers grow protozoa in defined or complex culture media under suitable temperature, pH, oxygen, and nutrient conditions, while aseptic techniques limit contamination and regular subculturing sustains viable populations. These cultures support investigations of cell structure, motility, metabolism, life cycles, and host–parasite interactions. They also provide standardized systems for testing antimicrobial compounds, examining disease mechanisms, and comparing responses to environmental changes, making in vitro culture valuable in microbiology, parasitology, ecology, and experimental biology.

We present procedures for demonstrating that ligands bind to the surface membrane of the cellulose-digesting protozoa in the gut of Formosan subterranean termites using fluorescent microscopy and that ligands coupled with lytic peptides kill these protozoa in vitro (anaerobic protozoa culture) and in vivo (injection into the termite hindgut).

Whole organ culture of the intervertebral disc (IVD) preserves the native extracellular matrix, cell phenotypes, and cellular-matrix interactions. Here we describe an IVD culture system using mouse lumbar and caudal IVDs in their functional spinal units and several applications utilizing this system.

In Vitro Culture and Differentiation of Primary Monocytes into Macrophages

This video demonstrates the priming of primary monocytes into macrophages. Primary human monocytes can be isolated and differentiated to an M1-like pro-inflammatory phenotype using the granulocyte-macrophage colony-stimulating factor (GM-CSF) or to an M2-like anti-inflammatory phenotype using the macrophage colony-stimulating factor (M-CSF).
