Rare Butterfly Conservation

Rare butterfly conservation is the science and practice of protecting threatened butterfly species, their habitats, and the ecological relationships that support their survival. It combines population monitoring with habitat protection and restoration, including preservation of host plants required by caterpillars and nectar resources used by adults; some programs also apply captive breeding and carefully planned reintroductions. In biology, these efforts help researchers understand population decline, genetic diversity, disease, and responses to climate change. Effective conservation can stabilize vulnerable populations, restore pollination and food-web interactions, and guide broader strategies for preserving biodiversity.

Here, we present a straightforward protocol for noninvasive genetic sampling of butterfly populations based on the field collection of residual egg debris. It can be used to confirm species identity and quantify genetic variation. This protocol can be easily adapted to broader groups for community science involvement.

In order to examine gene expression in the pupal wing tissue of Bicyclus anynana, we present an optimized protocol for in situ hybridizations using riboprobes. We also provide guidelines for the further optimization of this protocol for use in pupal wings of other Lepidopteran species.

Conservation of Momentum

Source: Nicholas Timmons, Asantha Cooray, PhD, Department of Physics & Astronomy, School of Physical Sciences, University of California, Irvine, CA The goal of this experiment is to test the concept of the conservation of momentum. By setting up a surface with very little friction, collisions between moving objects can be studied, including their initial and final momenta. The conservation of momentum is one of the most important laws in physics. When something is conserved in physics, the...

Here, we present protocols for 1) the laboratory captive propagation of the federally endangered Miami blue butterfly (Cyclargus thomasi bethunebakeri), and 2) assessing basic life history information such as immature development time and number of larval stadia. Both methods can be adapted for use with other ex situ conservation programs.

In Vivo Intracellular Recording of Photoreceptor Cell Spectral Sensitivity in a Butterfly

2025

This video demonstrates the procedure for in vivo intracellular recording of photoreceptor cells in the eye of a butterfly and determining the spectral sensitivity of individual cell types. This technique enables the study of sensitivities of individual photoreceptor cells to different light stimuli.
