Rat Tail Tendon

Rat tail tendon is a collagen-rich connective tissue that connects muscle to bone and transmits mechanical force, making it a useful model for studying tendon biology and connective-tissue mechanics. Its aligned collagen fibers and resident tenocytes form an extracellular matrix that bears tensile loads, while loading conditions influence matrix organization, cell behavior, and tissue remodeling. In biology research, rat tail tendon supports investigations of collagen structure, mechanical properties, wound healing, fibrosis, and mechanobiology. It is also used to evaluate biomaterials and tissue-engineering strategies aimed at restoring tendon function.

This article describes the experimental procedures used to prepare rat tail tendons for biomechanical and mechanobiological studies. Several features of the main steps in preparation are demonstrated, beginning with extraction, cross-sectional area measurement, rinsing and loading into the bioreactor chamber.

A Platelet-Rich Plasma Injection into the Sectioned Achilles Tendon of a Rat

This video demonstrates a technique for injecting platelet-rich plasma (PRP) into the sectioned Achilles tendon of rats. A small segment is excised from the Achilles tendon of a recipient rat, the site is sutured, and the rat is allowed to recover. Concurrently, whole blood is collected from a donor rat and centrifuged to obtain PRP, which is then activated and injected into the sutured site of the recipient rat to induce tendon repair.

Blood samples are useful for assessing biomarkers of physiological states or disease in vivo. Here we describe the methodology to sample blood from the lateral tail vein in the rat. This method provides rapid samples with minimal pain and invasiveness.

Here, we describe a simple tail vein blood sampling method in non-anesthetized rats using a vacuum extraction tube system. This method reduces the risk of direct exposure to blood and simplifies taking multiple samples from a single venipuncture.
