Proteasome Complex Characterization

Proteasome complex characterization is the systematic analysis of the composition, structure, assembly, and activity of proteasomes, ATP-dependent protease complexes that regulate protein turnover in cells. Researchers combine biochemical purification, native gel electrophoresis, mass spectrometry, structural imaging, and enzymatic assays to identify subunits, distinguish proteasome forms, and measure how ubiquitinated proteins enter the core particle and undergo controlled proteolysis. In biology, these approaches clarify how proteasomes maintain protein quality, influence cell-cycle progression and signaling, and respond to stress. Characterizing altered proteasome complexes also supports studies of neurodegeneration, cancer, immune regulation, and potential proteasome-targeted therapies.

The Proteasome

Eukaryotic cells can degrade proteins through several pathways. One of the most important amongst these is the ubiquitin-proteasome pathway. It helps the cell eliminate the misfolded, damaged, or unwarranted cytoplasmic proteins in a highly specific manner. In this pathway, the target proteins are first tagged with small proteins called ubiquitin. A series of enzymes carry out the ubiquitination of the target proteins - E1 (ubiquitin-activating enzyme), E2 (ubiquitin-conjugating enzyme), and E3...

Measuring Proteasome Activity in Different Subcellular Compartments of the Rat Brain

This video demonstrates how to measure proteasome activity in different subcellular compartments of the lateral amygdala from fear-conditioned and control rat brains using a fluorogenic peptide assay. The proteasome cleaves the fluorogenic peptide substrate, releasing fluorescent molecules, which are quantified using a plate reader to assess proteasome activity in the nuclear and synaptic protein fractions. Increased nuclear activity suggests changes in gene expression, while reduced synaptic...

Targeted protein degradation represents a major regulatory mechanism for cell function. It occurs via a conserved ubiquitin-proteasome pathway, which attaches polyubiquitin chains to the target protein that then serve as molecular “tags” for the 26S proteasome. Here, we describe a simple and reliable cell-free assay for proteasomal degradation of proteins.
