Renal Proximal Tubular Cells

Renal proximal tubular cells are specialized epithelial cells that reabsorb essential substances from the filtrate and help maintain the body’s fluid, electrolyte, and acid-base balance. Located in the proximal tubule of the nephron, they use transport proteins, ion gradients, and endocytic pathways to reclaim glucose, amino acids, bicarbonate, water, and other solutes while secreting selected waste products and medications. Their high metabolic activity and exposure to circulating compounds also make them vulnerable to ischemic and toxic injury. In biology and biomedical research, renal proximal tubular cells support studies of kidney function, drug-induced nephrotoxicity, transport mechanisms, disease processes, and potential protective therapies.

Here, we describe an in vitro protocol to model hypoxia/reoxygenation damage in the highly metabolic proximal tubular epithelial cells. This model is designed to induce metabolic stress-induced damage, which can be measured through the expression of proximal tubule damage markers and assessment of mitochondrial respiratory function.
