Reporter Ion Quantitation

Reporter ion quantitation is a mass spectrometry method that estimates the relative abundance of peptides or proteins by measuring diagnostic ions released from chemically labeled samples. In isobaric labeling workflows, such as tandem mass tag or iTRAQ analysis, sample-specific tags produce reporter ions during tandem mass spectrometric fragmentation; their intensities provide multiplexed abundance measurements after appropriate normalization and interference control. This approach enables simultaneous comparison of biological samples, supporting differential protein expression studies, pathway analysis, biomarker research, and characterization of cellular responses. Accurate reporter ion measurement links molecular changes to biological conditions while preserving efficient use of limited sample material.
