Ribosomal Proofreading

Ribosomal proofreading is the quality-control process that helps ribosomes select the correct transfer RNA (tRNA) during translation, preserving the accuracy of protein synthesis. As an aminoacyl-tRNA enters the decoding center, base pairing with the messenger RNA codon is tested through initial selection and a subsequent kinetic proofreading step; correct pairing promotes conformational changes and GTP hydrolysis by an elongation factor, whereas incorrect tRNAs are rejected before peptide-bond formation. This process limits mistranslation while maintaining the speed of translation, making it central to gene expression, studies of ribosome function, and research into how altered translational fidelity affects cellular physiology.
