Rrna Subtraction Protocol

An rRNA subtraction protocol is a molecular method that removes ribosomal RNA from a total RNA sample, enriching the messenger RNA and other non-ribosomal transcripts needed for transcriptome analysis. The procedure typically uses complementary oligonucleotide probes to hybridize with abundant rRNA molecules, followed by selective capture or enzymatic degradation of the probe-bound complexes while retaining the remaining RNA. By reducing rRNA dominance, this approach increases sequencing capacity for informative transcripts and supports RNA sequencing, gene-expression profiling, and studies of coding and noncoding RNA. It is particularly useful when analyzing degraded samples or transcriptomes in which polyadenylation-based selection is unsuitable.

This manuscript describes a detailed standardized protocol of high-throughput 16S rRNA-amplicon sequencing. The protocol introduces an integrated, uniformed, feasible, and inexpensive protocol starting from fecal sample collection through data analyses. This protocol enables analysis of large numbers of samples with rigorous standards and several controls.
