Secondary Antibody Staining

Secondary antibody staining is an immunodetection method that uses a labeled secondary antibody to visualize a target recognized by an unlabeled primary antibody, enabling sensitive detection in biological samples. The secondary antibody binds species-specific immunoglobulin regions, typically the Fc portion, while its fluorescent dye or enzyme label generates a signal after illumination or substrate addition. This two-step design can amplify detection because multiple secondary antibodies may associate with one primary antibody. In cell and tissue imaging, Western blotting, and related assays, secondary antibody staining helps identify protein localization, abundance, and changes across experimental conditions.
