Stem Loop Formation

Stem-loop formation is a nucleic acid secondary structure in which a single strand folds back on itself to create a double-stranded stem and an unpaired loop. It occurs when complementary sequences within the same RNA or DNA molecule align and form hydrogen-bonded base pairs, while intervening nucleotides remain unpaired; sequence, strand length, and solution conditions influence structural stability. In biology, stem-loops help RNA molecules fold into functional shapes and can regulate transcription termination, translation, splicing, and genome replication. They also support molecular biology methods, including primer design, hybridization assays, and prediction of RNA structure, making them important for studying gene regulation and nucleic acid function.

Zygotic Development And Stem Cell Formation

2023

The development of all multicellular organisms starts with the fusion of haploid cells called sperm and egg to form a diploid zygote. A zygote is a totipotent cell that can develop into a complete organism. The zygote undergoes cell division or cleavage to form an 8-cell mass. Until this stage, the cells are spherical, loosely attached, and remain totipotent. Totipotent cells are capable of developing both the embryonic and the extraembryonic tissues. However, as they continue to divide, they...

Rapid and Efficient Generation of Neurons from Human Pluripotent Stem Cells in a Multititre Plate Format

Protocols for neuronal differentiation of pluripotent human stem cells (hPSCs) are often time-consuming and require substantial cell culture skills. Here, we have adapted a small molecule-based differentiation procedure to a multititre plate format, allowing simple, rapid, and efficient generation of human neurons in a controlled manner.

We describe a novel surgical method for catheterizing 'intestinal loops' within the ileum of sheep. Once animals have recovered from surgery and have cleared antibiotics and analgesics, multiple treatments can be deposited directly in loops via the catheters.
