Stereo Microscope Guidance

Stereo microscope guidance provides practical instruction for using a stereo microscope, an optical instrument that produces a three-dimensional view of relatively large, opaque specimens at low magnification. It works through two separate optical paths that deliver slightly different views to each eye, while reflected illumination reveals surface features and adjustable zoom supports inspection at varying scales. In biology, effective guidance helps users position and orient specimens, select suitable lighting and magnification, and maintain depth perception during dissection, sorting, and manipulation. These skills support observations of insects, plant structures, tissues, and other specimens while improving handling precision and the quality of morphological data.

We describe here the use of a pH-sensitive green fluorescent protein variant, pHluorin, to study the spatio-temporal dynamics of axon guidance receptors trafficking at the cell surface. The pHluorin-tagged receptor is expressed both in cell culture and in vivo, using electroporation of the chick embryo.

This protocol describes a bioprinting methodology using an automated robotic depositing system that incorporates etched topographical guidance cues with the precision deposition of a cell bearing hydrogel bioink. The printed cells are directly delivered to the etched features and are able to sense and orientate with them.

We describe a new method for counting fishes, and estimating relative abundance (MaxN) and fish density using rotating stereo-video camera systems. We also demonstrate how to use distance from camera (Z distance) to estimate species-specific detectability.
