Intestinal Cell Line

An intestinal cell line is a population of cultured cells derived from intestinal tissue and maintained under controlled laboratory conditions to model intestinal structure and function. Depending on its origin and culture conditions, the cells can proliferate, differentiate into specialized epithelial phenotypes, form intercellular junctions, and develop barrier and transport properties that resemble aspects of the intestinal lining. Intestinal cell lines support studies of nutrient absorption, drug permeability, host-microbe interactions, inflammation, and intestinal disease in biology. They provide reproducible experimental systems that complement animal models and help researchers investigate epithelial responses, evaluate therapeutic compounds, and refine approaches to tissue engineering.

In this video protocol we give a step by step explanation of lentiviral transduction in organoids of primary intestinal epithelium and of processing and downstream analysis of these cultures by quantitative RT-PCR, RNA-microarray and immunohistochemistry.

Isolation and Characterization of Dendritic Cells and Macrophages from the Mouse Intestine

Here, we detail a methodology for the rapid isolation of mouse intestinal dendritic cells (DCs) and macrophages. Phenotypic characterization of intestinal DCs and macrophages is performed using multi-color flow cytometric analysis while magnetic bead enrichment followed by cell sorting is used to yield highly pure populations for functional studies.

Oral Delivery of Transformed Yeast Cells into the Mouse Intestinal Immune System

2025

This video showcases the oral administration of transformed yeast cells into a mouse model. The transformed yeast cells are orally administered using a gavage technique, reaching the mouse's stomach. Later, the cells migrate to the small intestine, where microfold cells engulf them and facilitate their transfer to Peyer's patches and gain access to the intestinal immune system.

Protocols for Analyzing the Role of Paneth Cells in Regenerating the Murine Intestine using Conditional Cre-lox Mouse Models

Intestinal epithelial stem cells (ISCs) are intermingled with Paneth cells. These cells are differentiated progeny of the ISC, which support the ISCs and provide antibacterial protection. Here we demonstrate how we used transgenic conditional mouse models to establish that Paneth cells play a crucial role in maintaining the intestinal epithelia.

Using Caco-2 Cells to Study Lipid Transport by the Intestine

Caco-2 cells can serve as an in vitro model to study the enterocyte transport of lipids, and lipid-soluble drugs/vitamins. The permeable membrane system separates the apical from the basolateral compartment, while the lentivirus expression system offers an effective gene overexpression method. The isolation of lipoproteins is confirmed by TEM.
