Stereo Microscopy

Stereo microscopy is an optical imaging technique that produces a three-dimensional view of specimens, making it useful for examining biological structures, surfaces, and spatial relationships. It uses two separate optical paths and slightly different viewing angles to deliver depth perception, typically at low magnification with reflected or transmitted illumination. In biology, stereo microscopes support specimen sorting, dissection, microinjection, and the observation of organisms, tissues, and developmental stages without extensive preparation. Their relatively large working distance also allows researchers to manipulate samples during imaging, providing a practical bridge between unaided observation and high-resolution compound microscopy.

We describe a new method for counting fishes, and estimating relative abundance (MaxN) and fish density using rotating stereo-video camera systems. We also demonstrate how to use distance from camera (Z distance) to estimate species-specific detectability.

This protocol uses multi-view stereo to generate three-dimensional (3D) models out of uncalibrated sequences of photographs, making it affordable and adjustable to a surgical setting. Strain maps between the 3D models are quantified with spline-based isogeometric kinematics, which facilitate representation of smooth surfaces over coarse meshes sharing the same parameterization.

Stereo-electroencephalography (SEEG) aids in localization of epileptogenic zones, however, remains relatively underutilized in the United States. The goal of this abstract is to provide a brief introduction to the technique of SEEG and further a detailed technique of using robotic assistance in the placement of SEEG electrodes.
