Sucrose Formulations

Sucrose formulations are prepared solutions or mixtures containing sucrose for controlled biological, biochemical, or laboratory use. Their effects depend on sucrose concentration and surrounding components: sucrose increases osmotic pressure, binds water, and can help stabilize membranes, proteins, and cells during handling, isolation, storage, or freezing. In biology, these formulations support cell and organelle fractionation, density-gradient separation, cryopreservation, and preparation of tissues or extracts, where consistent osmolarity and viscosity influence sample integrity and separation behavior. Selecting an appropriate concentration helps improve reproducibility while limiting osmotic stress and structural damage.

Freeze-drying is often an easy and convenient way to obtain dry products of viable bacterial cells. An issue of the process is cell survival. We detail here a procedure to investigate how cell survival during freeze-drying is influenced by the properties of the formulation used.
