Sulfite-reducing Bacteria

Sulfite-reducing bacteria are microorganisms that use sulfite as an electron acceptor during anaerobic respiration, making them important participants in the biological sulfur cycle and indicators of contamination in some environments. Under oxygen-limited conditions, sulfite reductase converts sulfite to sulfide, which may react with metals or detection reagents to produce a visible dark precipitate. Their growth and hydrogen sulfide production can be assessed in culture media to monitor food and water quality, investigate spoilage, and characterize microbial communities in sediments and wastewater. Studying these bacteria also helps clarify sulfur transformations and the environmental effects of anaerobic metabolism.

2025

Source: Anwar, Z., et al. Enrichment and Detection of Clostridium perfringens Toxinotypes in Retail Food Samples. J. Vis. Exp. (2019).In this video, sulfite-reducing bacteria are cultured in a sulfite-containing agar medium, where they grow anaerobically and reduce sulfite to hydrogen sulfide, forming black precipitates with iron ions. Black colonies are isolated from the agar depth using aseptic techniques and transferred to fresh medium for continued anaerobic incubation.

Source: Vy M. Dong and Daniel Kim, Department of Chemistry, University of California, Irvine, CA Controlling the reactivity and selectivity during the synthesis of a molecule is very important criteria for chemists. This has led to the development of many reagents that allow chemists to pick and choose reagents suitable for a given task. Quite often, a balance between reactivity and selectivity needs to be achieved. This experiment will use IR spectroscopy to monitor the reaction and to...

2015

Enhanced Reduced Representation Bisulfite Sequencing is a method for the preparation of sequencing libraries for DNA methylation analysis based on restriction enzyme digestion combined with cytosine bisulfite conversion. This protocol requires 50 ng of starting material and yields base pair resolution data at GC-rich genomic regions.

Reducing Line Loss

2024

In a three-phase circuit, line loss is an indicator of energy dissipated as heat due to the resistance of transmission lines. To address this, incorporating transformers into the system—a step-up transformer at the source and a step-down transformer at the load—is a strategic solution. Two three-phase transformers are introduced to improve this. With a step-up transformer at the source, the voltage is increased, thereby reducing the current in the transmission lines since power loss in...
