Threading Alignment

Threading alignment is a bioinformatics method that predicts a protein’s three-dimensional structure by fitting its amino acid sequence to experimentally determined structures of related or compatible proteins. Rather than relying only on sequence similarity, the method evaluates how well residues match the structural environment of a candidate template, including the arrangement of secondary-structure elements, conserved positions, and gaps or insertions. By identifying a plausible fold, threading alignment helps researchers model proteins with distant evolutionary relationships, annotate unknown sequences, and investigate molecular function. These models can support studies of protein evolution, interactions, disease-associated mutations, and structure-guided drug discovery.
