Transgenic Mouse Work

Transgenic mouse work is the genetic engineering of mice to carry an introduced DNA sequence, enabling researchers to examine how specific genes influence biology in a living mammal. In a typical approach, researchers introduce a DNA construct into a fertilized egg or modify embryonic stem cells, then generate mice in which the transgene becomes incorporated into the genome and may be inherited by offspring. These animals can reveal gene function, model human diseases, and support studies of development, physiology, and therapeutic responses. Careful breeding and molecular characterization are essential for linking observed traits to the introduced genetic change.

Source: Breid, S., et. al. Bioluminescence Imaging of Neuroinflammation in Transgenic Mice After Peripheral Inoculation of Alpha-Synuclein Fibrils. J. Vis. Exp. (2017)This video demonstrates the bioluminescence imaging of neuroinflammation in transgenic mice, showcasing luciferase-luciferin reactions in activated astrocytes. The protocol enables non-invasive quantification of astrocytic activation, aiding studies on CNS inflammation and therapeutic efficacy.

Imaging Transgenic Beta-Cells in an Immunodeficient Mouse Challenged with Diabetic Splenocytes

This video demonstrates an imaging-based method to visualize transplanted luciferase-expressing beta cells into a non-obese diabetic immunodeficient mouse. The introduction of splenocytes from a diabetic mouse leads to progressive beta cell destruction, visually confirmed by declining luminescence after substrate introduction.

Triggering and Monitoring Heat-Induced Seizures in a Transgenic Mouse Model

This video demonstrates the procedure to induce heat-induced seizures in a transgenic mouse with epilepsy-related gene mutations by gradually increasing body temperature in a controlled chamber. This protocol is used to study seizure mechanisms and potential therapies for epilepsy.

Ex Vivo Culture and Imaging of Oculomotor Slices from Transgenic Mouse Embryos

2025

This video demonstrates the ex vivo culture and imaging of oculomotor slices from transgenic mouse embryos. The embryos are isolated and embedded in agarose. Using a vibratome, slices containing the oculomotor nuclei and eyes are obtained. The slices are maintained in culture and imaged using a fluorescence microscope.

Culturing Fluorescent Excitatory Neurons from a Transgenic Mouse Pup Brain

2025

The video illustrates a technique for culturing fluorescent GABAergic and glutamatergic neurons from the cortex and hippocampal tissues of transgenic mouse pups. The process includes obtaining purified single-cell suspensions of neurons, sorting GABAergic neurons based on their fluorescence protein expression, and seeding them onto poly-L-lysine-coated plates to facilitate attachment and cell proliferation.
