Transwell Culture System

The Transwell Culture System is an in vitro platform that separates cells into upper and lower compartments with a porous membrane, enabling controlled study of cell behavior and intercellular communication. Cells can be cultured on one or both sides of the membrane, while defined pore sizes permit diffusion of soluble factors and, when appropriate, cell migration between compartments. In biology, Transwell assays help measure chemotaxis, membrane permeability, epithelial or endothelial barrier function, and interactions between different cell populations. By separating cells without eliminating biochemical communication, the system supports reproducible studies of tissue barriers, inflammation, cancer invasion, and other cellular processes.

The Transwell Migration Assay

2023

Cells migration in response to chemical cues is crucial to development, immunity and disease states such as cancer. To quantify cell migration, a simple assay was developed in 1961 by Dr. Stephen Boyden, which is now known as the transwell migration assay or Boyden chamber assay. This set-up consists an insert which separates the wells of a multiwell plate into top and bottom compartments. Cells whose migration is to be studied are seeded into the top compartment and the chemoattractant...

Interstitial fluid flow is elevated in solid tumors and can modulate tumor cell invasion. Here we describe a technique to apply interstitial fluid flow to cells embedded in a matrix and then measure its effects on cell invasion. This technique can be easily adapted to study other systems.

A Technique to Establish a Bacterial Infection Model in an Ex Vivo Organ Culture

2025

This video demonstrates a method to establish an infection model in placental tissues to study infection propagation. An ex vivo placental organ culture is infected with Listeria monocytogenes, a facultative intracellular parasite. The bacteria invade extravillous trophoblasts (EVTs) to enter the placental tissue and then propagate to other cells via cell-to-cell spread.
