Triglycerides

Triglycerides are lipids formed from one glycerol molecule and three fatty acids, serving as a major form of energy storage in animals and plants. Cells assemble them by esterifying fatty acids to glycerol and store them mainly in lipid droplets; when energy is needed, lipases hydrolyze triglycerides, releasing fatty acids for oxidation and glycerol for metabolic pathways. In biology, triglycerides help explain how organisms balance energy intake, storage, and use, while their transport in lipoproteins connects lipid metabolism to digestion and circulation. Studying triglyceride synthesis and breakdown supports research on nutrition, obesity, diabetes, cardiovascular disease, and metabolic regulation.

Here we describe a technique to cannulate the mesenteric lymph duct in rats which enables quantification of lipid and drug transport via the lymphatic system following intestinal delivery. The technique can be adapted to assess mesenteric lymph concentrations and/or transport of fluid, immune cells, peptides, proteins and lipophilic molecules.

Mammalian integument contains solvent-extractable lipids that can provide chemical compositions characteristic of individual species. This paper presents a routine method for separating broad lipid classes isolated from integumentary tissues using thin layer chromatography and determining the triacylglyceride profile by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry.
