Tumor Transplantation

Tumor transplantation is the experimental transfer of tumor cells or tissue into a living recipient to study cancer growth, host responses, and treatment effects. The transplanted material must survive, engraft, and interact with its new tissue environment, with outcomes influenced by donor-recipient compatibility, immune activity, and the tumor microenvironment. In biology, syngeneic, allogeneic, and xenograft models help researchers investigate tumor progression, metastasis, and interactions between cancer and host tissues. These models also support preclinical evaluation of anticancer drugs and immunotherapies, although differences between experimental hosts and patients can limit how directly findings translate to clinical care.

The video explains a stepwise surgical method for orthotopic implantation of a tumor fragment from a cancerous donor to the pancreas of a non-cancerous recipient mice for development of pancreatic cancer. Orthotopically implanted PDAC could recapture the tissue microenvironment of human pancreatic cancers than conventional subcutaneous (SC) models more accurately.

The transplantation of cancer cells is an important tool for the identification of cancer mechanisms and therapeutic responses. Current techniques depend on immune-incompetent animals. Here, we describe a method to transplant zebrafish tumor cells into immune-competent embryos for the long-term analysis of tumor cell behavior and in vivo drug responses.

Limiting dilution cell transplantation assays are used to determine the frequency of tumor-propagating cells. This protocol describes a method for generating syngeneic zebrafish that develop fluorescently-labeled leukemia and details how to isolate and transplant these leukemia cells at limiting dilution into the peritoneal cavity of adult zebrafish.
