Xenograft Drug Testing

Xenograft drug testing is a preclinical method that evaluates how candidate therapies affect human cells or tissues transplanted into a living animal, commonly to model cancer treatment responses. In a typical study, human tumor cells or tissue fragments are implanted into immunodeficient mice, allowing the graft to grow before researchers administer a drug and compare tumor growth, regression, or survival with untreated controls. This approach helps assess therapeutic efficacy, dose response, and potential drug resistance before clinical studies. Because the host lacks a fully functioning immune system, xenografts provide valuable biological context while requiring complementary models to evaluate immune-mediated effects.

Protocols for germ cell transplantation and testis tissue xenografting are described. Germ cell transplantation results in donor-derived spermatogenesis in recipient testes and represents a functional reconstitution assay for identification of spermatogonial stem cells (SSCs). Testis tissue xenografting reproduces testis development and spermatogenesis of various donor species in recipient mice.

Preclinical models aim to advance the knowledge of cancer biology and predict treatment efficacy. This paper describes the generation of zebrafish-based patient-derived xenografts (zPDXs) with tumor tissue fragments. The zPDXs were treated with chemotherapy, the therapeutic effect of which was assessed in terms of cell apoptosis of the transplanted tissue.
