Z Stack Analysis

Z stack analysis is a microscopy approach that examines a series of images acquired at successive focal planes through a biological specimen, allowing researchers to study structures across its depth. During imaging, the microscope changes the focal position in defined z-axis increments and records each optical section; software can then align, segment, and reconstruct these planes into a three-dimensional representation. In biology, Z stack analysis helps quantify cell morphology, tissue organization, protein localization, and interactions within complex specimens. It is particularly useful for fluorescence and confocal microscopy, where depth-resolved imaging improves interpretation compared with a single two-dimensional focal plane.

Scaling of Engineered Vascular Grafts Using 3D Printed Guides and the Ring Stacking Method

Scalable engineered blood vessels would improve clinical applicability. Using easily sizable 3D-printed guides, rings of vascular smooth muscle were created and stacked into a tubular form, forming a vascular graft. Grafts can be sized to meet the range of human coronary artery dimensions by simply changing the 3D-printed guide size.

Here we provide a detailed procedure for large-scale production of research-grade AAV vectors using adherent HEK 293 cells grown in cell stacks and affinity chromatography purification. This protocol consistently yields >1 x 1013 vector genomes/mL, providing vector quantities appropriate for large animal studies.

The key steps of living anionic polymerization of phenyl glycidyl ether (PheGE) on methoxy-polyethylene glycol (mPEG-b-PPheGE) are described. The resulting block copolymer micelles (BCMs) were loaded with doxorubicin 14% (wt%) and sustained release of drug over 4 days under physiologically relevant conditions was obtained.

Clock Scan Protocol for Image Analysis: ImageJ Plugins

This paper describes two novel ImageJ plugins for 'Clock Scan' image analysis. These plugins expand the functionality of the original visual basic 6 program and, most importantly, make the program available to a large research community by bundling it with the ImageJ free image analysis software package.
