Zip-code Regions

Zip-code regions are specific nucleotide sequences, often found in messenger RNA (mRNA) untranslated regions, that direct transcripts to particular locations within a cell. RNA-binding proteins recognize these cis-acting signals and, together with transport machinery and the cytoskeleton, deliver the associated mRNAs to defined subcellular compartments, where local translation can occur. In biology, this spatial control helps establish cell polarity, supports neuronal development, and coordinates protein production during embryogenesis and tissue organization. Studying zip-code regions clarifies how cells regulate gene expression beyond transcription and can reveal mechanisms disrupted in developmental disorders or disease.

In this manuscript, a protocol is presented to perform FACS-based measurement of BK-polyomavirus transcriptional activity by using HEK293T cells transfected with a bidirectional reporter plasmid expressing tdTomato and eGFP. This method further allows to quantitatively determine the influence of novel compounds on viral transcription.

New Methods to Study Gustatory Coding

We present three new methods to study gustatory coding. Using a simple animal, the moth Manduca sexta (Manduca), we describe a dissection protocol, the use of extracellular tetrodes to record the activity of multiple gustatory receptor neurons, and a system for delivering and monitoring precisely timed pulses of tastants.
